The Subway store on Bridge Street has permanently closed after its lease expired.
It is not known how many jobs were at risk but the sandwich company says it is trying to move the workers to other stores across town.
The store has already had its equipment removed and some signage taken down.
A spokesperson for Subway said: "We can confirm that the Subway store at 59/60 Bridge Street, Swindon has closed permanently due to the expiry of its lease.
"The franchise owner hopes to transfer the Sandwiches Artists employed at the Bridge Street store across to his four other stores in Swindon, where possible.
"Guests can find these stores at Brunel Shopping Centre, Hobley Drive, 9 Regent Circus and Hoopers Place Old Town."
