The A419 southbound has been completely shut for roadworks that will last the whole weekend.
The road towards Swindon is closed from the B4696 Spine Road East to the B4040 and will reopen in the early hours of Monday morning.
The AA has said that delays are expected.
There is a detour in operation via the A4696 through Ashton Keynes followed by the B4040 to Cricklade.
Inbound Stagecoach West bus routes have also been affected, and will not stop at Latton.
Stagecoach West said in a statement: “Due to the weekend closure of the A419, we will be diverting via Ashton Keynes to Cricklade and then via to Purton Stoke.
"During this time we will be unable to service Latton. Sorry for any inconvenience.”
